Your sample is processed in a certified lab, turning your DNA into the genetic information used to generate your 3X4 / GENEFIT report. Processing at the lab usually takes about 3-4 weeks.
Sample arrives at the lab
The lab visually inspects your cheek sample vial and scans the kit code to check it in for processing.
If any registration problems are found (e.g., an unregistered kit), our operations team is notified to assist.
DNA extraction
Extraction buffer is added to your sample, which breaks open the cells to release the DNA.
The sample is then “amplified”, which increases the amount of DNA in the sample.
The final extraction step is to break up and concentrate the DNA strands, in a process called fragmentation and precipitation.
Genotyping
Your DNA sample is run on 3X4’s custom SNP microarray, a tiny chip designed to scan thousands of genetic markers and determine your gene variants.
The array contains microscopic probes that act as templates, for each gene variant. The matching DNA in your sample will bind to the probes on the array. At the probe location, a fluorescent signal is emitted, which allows the gene variant to be identified.
Genotyping is performed on the GeneTitanTM machine, a machine that captures the fluorescent signals. The resulting data is analyzed and quality checked using the Axiom Analysis Software Suite to generate your genetic dataset.
About 3X4’s Custom SNP Micro-array
The 3X4 science team worked with Thermo Fisher Scientific to design a custom microarray. The customized array was internally and externally validated in collaboration with Thermo Fischer Scientific and partner laboratories to confirm analytical accuracy and fitness for purpose.
3X4 Quality Control
Quality control is performed both on the sample and the individual probe sets.
For the DNA sample, we assess the overall call rate and sample purity.
For the genetic variants, the individual cluster quality for the genes covered by our reports are confirmed to meet quality standards, before we generate your 3X4 / GENEFIT report.
